Cell Tracker Dye

Cell tracker dyes are fluorescent probes used to label living cells so their location, movement, survival, or lineage can be followed over time. They typically enter cells in a membrane-permeable form, then undergo intracellular conversion or binding that produces a retained fluorescent signal, allowing labeled cells to be visualized by fluorescence microscopy or flow cytometry. In biology, these dyes support studies of cell migration, proliferation, differentiation, and interactions within cultures or tissues. Because signal inheritance and dilution can reveal cell division, cell tracker dyes provide a practical way to connect cellular behavior with developmental, immunological, and disease-related processes.

We recently proposed a method that allows dichoptic visual stimulus presentation and binocular eye tracking simultaneously1. The key is the combination of an infrared eye tracker and the corresponding infrared-transparent mirrors. This manuscript provides an in depth protocol for initial setup and everyday operation.

Successful use of cell tracking dyes to monitor immune cell function and proliferation involves several critical steps. We describe methods for: 1) obtaining bright, uniform, reproducible label-ing with membrane dyes; 2) selecting fluorochromes and data acquisition conditions; and 3) choosing a model to quantify cell proliferation based on dye dilution.
