Flow Cytometric Sorting

Flow cytometric sorting is a method that analyzes and separates individual cells according to measurable features such as size, granularity, and fluorescence, enabling researchers to isolate defined cell populations from a heterogeneous sample. As cells pass in a fluid stream through laser beams, detectors record their light-scattering and fluorescent signals; selected cells are then enclosed in droplets and separated by electrostatic deflection into collection vessels. In biology, this technique supports cell-type purification, immune-cell profiling, stem-cell research, gene expression studies, and downstream molecular or functional assays. By combining multiparameter analysis with physical isolation, flow cytometric sorting links cellular phenotype to precise experimental investigation.

There is growing interest in the quantitative characterization of intestinal lymphocytes owing to increasing recognition that these cells play a critical role in a variety of intestinal and systemic diseases. In this protocol, we describe how to isolate single cell populations from different small-intestinal compartments for subsequent flow cytometric characterization.

We describe a sorting strategy for mouse spermatids using flow cytometry. Spermatids are sorted into four highly pure populations, including round (spermiogenesis steps 1-9), early elongating (spermiogenesis steps 10-12), late elongating (spermiogenesis steps 13-14) and elongated spermatids (spermiogenesis steps 15-16). DNA staining, size and granulosity are used as selection parameters.
