Closed Complex

A closed complex is an early molecular assembly in which a protein or enzyme binds its nucleic-acid target while the DNA or RNA remains in its original, unseparated structure. In bacterial transcription, RNA polymerase recognizes promoter elements with help from a sigma factor, forming a closed complex before locally melting promoter DNA to produce the open complex and initiate RNA synthesis. Studying this transition clarifies how cells control gene expression, how promoter sequence and regulatory proteins influence transcription, and how mutations or antimicrobial compounds can alter transcription initiation. The concept is also useful for interpreting structural and biochemical experiments on nucleic-acid–protein interactions.
