Collagen Fiber Imaging

Collagen fiber imaging is the visualization and measurement of collagen’s three-dimensional organization, orientation, density, and remodeling in biological tissues. In methods such as second-harmonic generation microscopy, the non-centrosymmetric, highly ordered structure of collagen generates a label-free optical signal when illuminated with pulsed laser light, allowing fibers to be resolved without staining; fluorescence and electron microscopy provide complementary molecular or ultrastructural information. These approaches help researchers relate extracellular-matrix architecture to tissue mechanics, development, wound healing, fibrosis, and tumor progression. Quantitative analysis of fiber alignment, thickness, and network changes can reveal how cells interact with their matrix and identify structural signatures of disease.

This procedure demonstrates in vivo near IR fluorescence imaging of collagen remodeling activities in mice as well as ex vivo staining of collagens in tissue sections using caged collagen mimetic peptides that can be photo-triggered to hybridize with denatured collagen strands.

This protocol demonstrates the use of a microfluidic channel with changing geometry along the fluid flow direction to generate extensional strain (stretching) to align fibers in a 3D collagen hydrogel (<250 µm in thickness). The resulting alignment extends across several millimeters and is influenced by the extensional strain rate.
