Core Promoter

A core promoter is the minimal DNA region surrounding a gene’s transcription start site that directs assembly of the basal transcription machinery and determines where transcription begins. In eukaryotes, sequence elements such as the TATA box, initiator (Inr), downstream promoter element, or TFIIB recognition element can be recognized by general transcription factors, helping recruit RNA polymerase II and position it for RNA synthesis. Core promoters work with regulatory elements such as enhancers to control gene expression in different cell types and conditions. Their study supports analysis of transcriptional regulation, reporter-gene assays, disease-associated mutations, and the design of synthetic gene-expression systems.
