Cryoprotectant Toxicity Reduction

Cryoprotectant toxicity reduction is the systematic control of chemical injury caused by cryoprotective agents during biological cryopreservation. These compounds protect cells by limiting ice formation and reducing dehydration, but high concentrations or prolonged exposure can disrupt membranes, alter proteins, and impair viability; toxicity reduction therefore relies on balancing dose, exposure time, temperature, and loading or removal rates. Strategies may include stepwise addition and dilution, optimized cooling and warming conditions, and substitution with less toxic cryoprotectants. Improving this balance supports the recovery of viable cells, tissues, embryos, and other biological samples for research, clinical preservation, and biobanking.

Liver macrophages, named Kupffer cells, are responsible for the capture of circulating nanoparticles. We describe here a method, of high cell purity and yield, for Kupffer cell isolation. The modified LDH assay is used here to measure the toxicity induced by carbon nanotubes in Kupffer cells.
