Dead Cell Inhibition

Dead cell inhibition is a biological phenomenon in which nonviable cells or their remnants reduce the survival, growth, or activity of nearby living cells. It can occur when dying cells release intracellular contents, damage-associated signals, or other soluble factors that alter the local environment and trigger stress, inflammation, or growth arrest in neighboring cells. Studying this interaction helps researchers distinguish direct effects of cell death from secondary responses in tissues and cell cultures. The concept is relevant to tissue injury, infection, cancer biology, and regenerative research, where the accumulation and clearance of dead cells can influence inflammation, recovery, and experimental outcomes.

Feedback Inhibition

Biochemical reactions are occurring constantly in cells, converting starting substances to different products, usually with the help of enzymes that speed the reactions. Without enzymes, it would take far too long for most reactions to occur to be useful to the cell! Since enzymes help control the rate of reactions, their activity is regulated so that appropriate amounts of starting materials, intermediate metabolites, and products are maintained in the cell. Excessive build-up or depletion of...

Neurodegenerative diseases are associated with dysregulated microglia functions. This article outlines an in vitro assay of phagocytosis of neuroblastoma cells by iPSC-macrophages. Quantitative microscopy readouts are described for both live-cell time-lapse imaging and fixed-cell high-content imaging.

Habituation and prepulse inhibition of startle are operational measures of sensory gating. Sensory gating is disrupted in schizophrenia, and some other mental disorders and neurodegenerative diseases. We here describe a standard protocol to assess short-term and long-term habituation as well as prepulse inhibition of acoustic startle responses in rats and mice.

Immunoblot Analysis of Apoptosis Inhibition in Host Cells Expressing an Antiapoptotic Bacterial Protein

Source: Berens, C. et al. Applying an Inducible Expression System to Study Interference of Bacterial Virulence Factors with Intracellular Signaling. J. Vis. Exp. (2015)This video demonstrates the use of immunoblotting to assess apoptosis inhibition by the bacterial effector protein CaeB in human cells. It outlines the steps for detecting apoptotic markers following doxycycline-induced expression of pro-apoptotic proteins in host cells expressing the effector.
