Protease Digestion Measurement

Protease digestion measurement is the quantitative assessment of how efficiently proteolytic enzymes break down proteins, an essential approach for studying protein turnover, digestion, and enzyme function in biology. Proteases hydrolyze peptide bonds, and researchers measure digestion by tracking changes in protein or substrate concentration, the release of peptides or amino acids, or the formation of products over time under defined conditions such as pH, temperature, and enzyme concentration. These measurements help determine reaction rates, enzyme activity, substrate specificity, and the effects of inhibitors or environmental changes. The approach supports research in cellular regulation, physiology, biotechnology, and disease mechanisms.

Innate defenses to virus infections are triggered by pattern recognition receptors (PRRs). The two cytoplasmic PRRs RIG-I and PKR bind to viral signature RNAs, change conformation, oligomerize, and activate antiviral signaling. Methods are described which allow to conveniently monitor the conformational switching and the oligomerization of these cytoplasmic PRRs.

The Protease Fluorescent Detection Kit is designed for the measurement of protease activity using fluorometry. It is also suitable for detection of trace amounts of protease contamination. The method is based on the proteolytic hydroysis of a proprietary formulation of a FITC-labeled casein substrate.

This video demonstrates an in vitro assay to quantify neutrophil serine protease activity in sputum samples. The sample containing secreted protease is incubated with a fluorescent reporter bearing a recognition motif. Cleavage of the motif by the protease enables individual fluorescence emission by the donor and acceptor fluorophore of the reporter, indicating protease activity in the sample.
