Disease Modeling

Disease modeling is the use of experimental systems and computational approaches to represent how diseases arise, progress, and respond to intervention. Models recreate selected features of human disease by combining genetic, cellular, tissue, organismal, or mathematical representations with controlled perturbations, measurements, and comparisons to healthy states. In biology, researchers use cell cultures, organoids, animal models, and simulations to examine disease mechanisms, identify biomarkers, and test candidate therapies before clinical studies. Because each model captures only part of disease complexity, integrating complementary models can improve biological interpretation, reveal therapeutic targets, and support more predictive and ethically informed research.

The inoculation of Trypanosoma cruzi in fertile eggs prior to incubation renders the parasite kDNA minicircle integration in embryo cells genome. Crossbreeding reveals the vertical transfer of the mutations to progeny. The kDNA integrates into coding regions at several chromosomes and the chickens die with an inflammatory autoimmune heart disease.

One of the pathological characteristics of AD is the formation of Amyloid β protein positive neuritic plaques. In this protocol we describe two methods to detect neuritic plaques in transgenic AD model mice: immunohistochemical detection using the ABC and DAB method and fluorescent detection using thioflavin S staining method.

In Parkinson's disease and movement disorders in general, sensitive and reliable behavioral assays are essential for testing novel potential therapeutics. Here, we describe a manageable battery of sensorimotor tests for mice that are sensitive to varying degrees of injury to the nigrostriatal system and useful for preclinical studies.
