Drug Assessment

Drug assessment is the systematic evaluation of a drug’s biological activity, effectiveness, quality, and safety before or during its use in research and medicine. It combines molecular, cellular, and organism-level studies to determine how a compound interacts with its target, alters biological pathways, is absorbed and distributed, and produces beneficial or adverse responses under defined conditions. In biology, these assessments may use biochemical assays, cell-based models, and animal or clinical studies to compare dose-dependent effects and identify toxicity. Reliable drug assessment supports candidate selection, mechanism-of-action research, risk evaluation, and the development of treatments that provide greater therapeutic benefit with fewer harmful effects.

Drug self-administration and ultrasonic vocalizations (USV) are used as behavioral assessments in animal research, but rarely in combination. The purpose of this article is to describe the advantages of recording USVs during drug self-administration procedures to assess affective responses to drug experience.

The Mesenteric Lymph Duct Cannulated Rat Model: Application to the Assessment of Intestinal Lymphatic Drug Transport

Here we describe a technique to cannulate the mesenteric lymph duct in rats which enables quantification of lipid and drug transport via the lymphatic system following intestinal delivery. The technique can be adapted to assess mesenteric lymph concentrations and/or transport of fluid, immune cells, peptides, proteins and lipophilic molecules.

We exposed a microphysiological system (MPS) with intestine and liver organoids to acetaminophen (APAP). This article describes the methods for organoid production and APAP pharmacokinetic and toxicological property assessments in the MPS. It also describes the tissue functionality analyses necessary to validate the results.
