E. Coli Extract System

E. coli extract systems are cell-free biological platforms that use components derived from Escherichia coli to carry out biochemical reactions outside living cells. An E. coli lysate supplies ribosomes, enzymes, transfer RNAs, energy-regeneration components, and other molecular machinery that can transcribe DNA into RNA and translate RNA into protein when provided with a template and appropriate substrates. These systems support rapid protein production, analysis of gene expression, enzyme and pathway reconstruction, and testing of synthetic biology designs under controlled conditions. Because they avoid cellular growth constraints, E. coli extracts also help researchers investigate molecular mechanisms and prototype biological circuits efficiently.

Protein co-expression is a powerful alternative to the reconstitution in vitro of protein complexes, and is of help in performing biochemical and genetic tests in vivo. Here we report on the use of protein co-expression in Escherichia coli to obtain protein complexes, and to tune the mutation frequency of cells.
