Egfp Overexpression

EGFP overexpression is the deliberate production of enhanced green fluorescent protein at levels above those normally present in a biological system, allowing researchers to visualize gene expression, protein localization, and cell behavior. Typically, an EGFP coding sequence is introduced into cells on an expression construct controlled by a promoter; transcription and translation generate abundant fluorescent protein, whose chromophore forms within the protein and emits green light after excitation. This approach supports live-cell imaging, reporter assays, lineage tracing, and evaluation of transfection or gene-regulation systems in biology. Because fluorescence can be measured without destroying cells, EGFP overexpression helps connect molecular activity with cellular outcomes, although excessive expression may perturb normal cell physiology.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
