Embryo Exteriorization

Embryo exteriorization is a surgical technique that temporarily brings an embryo, or the embryonic tissue containing it, outside the body for direct observation, manipulation, or experimental analysis. In developmental biology, researchers carefully expose the uterus or embryonic region under anesthesia, maintain tissue hydration and temperature, and return it promptly to preserve normal development. This approach enables procedures such as microsurgery, labeling, imaging, gene delivery, and localized treatment while the embryo remains connected to maternal support. By providing access to otherwise enclosed stages of development, embryo exteriorization helps clarify tissue formation, cell movements, and developmental responses in vivo.

This article describes a simple model for stimulating angiogenesis in the rat mesentery. The model produces dramatic increases in capillary sprouting, vascular area and vascular density over a relatively short time course in a tissue that allows en face visualization of entire microvascular networks down to the single cell level.

Here, we present a protocol to inject ultrasound microbubble contrast agents into living, isolated late-gestation stage murine embryos. This method enables the study of perfusion parameters and of vascular molecular markers within the embryo using contrast-enhanced high-frequency ultrasound imaging.

Microinjection of Live Drosophila Embryos: Early Delivery of Reagents to the Developing Embryo

2023

This video describes how to perform microinjections that deliver reagents into live Drosophila embryos. Researchers can use this method to introduce exogenous compounds like nucleic acids and proteins during the early stages of embryo development. The featured protocol demonstrates how to set up and perform the procedure for injection of any soluble reagent–here, a fluorescent protein for live imaging experiments.
