Endogenous Promoter Expression

Endogenous promoter expression is the production of a gene’s RNA under the control of its native promoter, preserving the regulatory context found in the organism or cell. This process occurs when transcription factors and other regulatory proteins interact with promoter DNA, while chromatin structure and cellular signals influence whether RNA polymerase initiates transcription. Studying endogenous promoter expression helps researchers measure natural gene activity, examine cell-type-specific and condition-dependent regulation, and understand how genetic changes affect transcription. It also supports the design and interpretation of reporter assays, gene-editing experiments, and models of development, disease, and cellular response.

We provide a method for testing BRCA1 variants in a tissue culture based assay for homologous recombination repair of DNA damage by depleting endogenous BRCA1 protein from a cell using RNAi and replacing it with a BRCA1 point mutant that contains a coding change.

Herein we present a protocol to identify, isolate, and clone a plant promoter. Further, we describe the characterization of the nodule-specific promoter in the common bean hairy roots.
