Epidermal Renewal

Epidermal renewal is the biological process through which the outer layer of skin continually replaces cells lost through normal shedding, maintaining the protective barrier between the body and its environment. In the basal layer, epidermal stem and progenitor cells divide to produce keratinocytes that migrate outward, differentiate, accumulate keratin, and eventually detach from the surface in a process called desquamation. This coordinated cycle supports barrier integrity, hydration, and defense against physical and microbial stress. Studying epidermal renewal helps explain wound healing, skin aging, inflammatory disorders, and the effects of drugs or environmental exposures on tissue maintenance.

The skin is divided into epidermis, dermis, and hypodermis, the skin's outermost, middle, and inner layers. SC1 functions through dual inhibition of Ras- GAP and ERK1. We tested the function of SC1 in supporting mouse ES cell self-renewal in the absence of LIF and showed that SC1 is able to maintain self-renewal of mouse ES cell cultures.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

Pharmacologic Induction of Epidermal Melanin and Protection Against Sunburn in a Humanized Mouse Model

Epidermal melanin is induced by topical application of forskolin in a murine model of the fair-skinned UV-sensitive human. Pharmacologic manipulation of cAMP levels in the skin and epidermal darkening strongly protect against UV-mediated inflammation (sunburn) as measured by the minimum erythematous dose (MED) assay.
