Erythropoiesis

Erythropoiesis is the biological process that produces red blood cells, or erythrocytes, from hematopoietic stem and progenitor cells, enabling oxygen transport throughout the body. In adults, it occurs mainly in bone marrow as progenitors undergo lineage commitment, proliferation, hemoglobin accumulation, nuclear extrusion, and maturation into reticulocytes; reduced oxygen availability stimulates renal erythropoietin, which promotes these stages. Studying erythropoiesis helps explain normal blood physiology and disorders such as anemia, while supporting research into stem cell differentiation, erythropoietin-based therapies, blood production, and the cellular effects of disease or environmental stress.

An ex vivo protocol to generate mature human red blood cells from hematopoietic stem/progenitors is described. Additionally we describe an efficient lentiviral-delivery method to knockdown the transcription factor TAL1 in primary erythroid cells. The efficiency of lentivirus mediated gene delivery is demonstrated using GFP expressing viruses.
