Exosome Mirna Profiling

Exosome miRNA profiling is a molecular analysis method used to identify and quantify microRNAs carried within exosomes, small extracellular vesicles that can reflect cellular activity and communication. The workflow typically isolates exosomes from biofluids or cell cultures, breaks open their membranes, extracts the enclosed RNA, and measures miRNA abundance using reverse-transcription quantitative PCR, microarrays, or next-generation sequencing. By comparing miRNA profiles across samples, researchers can investigate intercellular signaling, disease-associated molecular changes, and potential biomarkers. In biology, this approach supports studies of development, cancer, immune responses, and therapeutic communication while helping clarify how extracellular vesicles influence recipient cells.

The presence of stable microRNAs (miRNAs) in exosomes has generated immense interest as a novel mode of intercellular communication, for their potential utility as biomarkers and as a route for therapeutic intervention. Here we demonstrate exosome purification from blood and culture media followed by quantitative PCR to identify miRNAs being transported.

This paper demonstrates methods for the isolation, purification and detection of exosomes, as well as techniques for analysis of their molecular content. These methods are adaptable for exosome isolation from both cell culture media and biological fluids, and can beyond analysis of molecular content also be useful in functional studies.
