Expansion Medium

Expansion medium is a specialized cell culture formulation designed to support the growth and increase of cell populations under controlled laboratory conditions. It provides essential nutrients, salts, buffering components, and, when required, growth-promoting factors that help maintain cell viability and stimulate proliferation while cells remain in an appropriate physiological environment. Researchers use expansion media to generate sufficient numbers of cells for biological studies, disease modeling, drug testing, tissue engineering, and regenerative medicine. By adjusting the medium to the cell type and culture objective, investigators can influence expansion efficiency, phenotype maintenance, and the quality of cells produced for downstream experiments.

This video demonstrates the production of oligodendrocyte-conditioned medium (OCM) from oligodendrocyte lineage cells by stressing them to enhance their molecular release into the medium. This medium, which is rich in growth factors, cytokines, and other bioactive molecules, can be added to neuron cultures to gain insight into the impact of oligodendrocytes-secreted factors on neuronal physiology and connectivity.

Expansion Microscopy: High-Resolution Fluorescent Imaging with a Conventional Microscope

Here, we present a detailed protocol combining cryo-fixation and expansion microscopy methods (Cryo-ExM), allowing for high-resolution imaging with structural preservation adapted to various biological samples. Biological samples are frozen, embedded in a swellable polymer, denatured, expanded, and immunolabeled, enabling super-resolution imaging with standard light microscopes.
