Syto 9 Staining

SYTO 9 staining is a fluorescence-based method for labeling nucleic acids in cells, making microorganisms visible for imaging, counting, and viability analysis. As a membrane-permeable green fluorescent dye, SYTO 9 enters cells and binds DNA and RNA, increasing its fluorescence; in bacterial viability assays, it is often combined with propidium iodide, which enters cells with compromised membranes and can alter or quench SYTO 9 fluorescence. This differential staining helps distinguish membrane-intact from membrane-damaged bacteria in mixed populations. The method supports studies of microbial growth, biofilms, antimicrobial effects, and environmental samples, while fluorescence microscopy and flow cytometry provide quantitative or spatial measurements.
