Filter Aided Sample Preparation

Filter Aided Sample Preparation (FASP) is a protein-processing technique that prepares complex biological samples for mass spectrometry by combining detergent removal, buffer exchange, and enzymatic digestion in an ultrafiltration device. Proteins are retained by a molecular-weight cutoff membrane while detergents and other low-molecular-weight contaminants pass through during centrifugation; the retained proteins are then reduced, alkylated, and digested, commonly with trypsin, on the filter. FASP supports proteomic analysis of tissues, cells, and fluids, improving compatibility with liquid chromatography–tandem mass spectrometry and enabling identification and relative quantification of proteins in challenging biological samples.

We describe a protocol for filtration of water samples with a filter cartridge and extraction of environmental DNA (eDNA) without having to cut open the housing to remove the filter. This protocol is developed for metabarcoding eDNA from fishes, but is also applicable to eDNA from other organisms.

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Sample preparation is the way in which a sample is treated to prepare for analysis. Careful sample preparation is critical in analytical chemistry to accurately generate either a standard or unknown sample for a chemical measurement. Errors in analytical chemistry methods are categorized as random or systematic. Random errors are errors due to change and are often due to noise in instrument. Systematic errors are due to...
