Fm 1-43 Dye

FM1-43 dye is a fluorescent amphipathic styryl dye used to visualize plasma-membrane dynamics and vesicle recycling in living cells. It remains weakly fluorescent in aqueous solution but becomes strongly fluorescent when inserted into lipid bilayers, allowing membrane internalization and dye release to be monitored. In neurons and other secretory cells, researchers use FM1-43 to follow endocytosis, label synaptic or secretory vesicles, and measure exocytosis through activity-dependent changes in fluorescence. This approach provides a readout of membrane trafficking, helping connect cellular stimulation with vesicle turnover and secretion.

Here, we describe a S. pneumoniae serotype 1 strain 519/43 that can be genetically modified by using its ability to naturally acquire DNA and a suicide-plasmid. As proof of principle, an isogenic mutant in the pneumolysin (ply) gene was made.

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.

In this video, Drosophila larvae nerve fibers are immersed in FM dye, which binds to cell membrane lipids. Suction microelectrode and electrical pulses stimulate the endocytosis process, incorporating the FM dye into newly formed endocytic vesicles. This method, which visualizes presynaptic stages of neurotransmission, is useful for studying synaptic function and neurotransmitter release.
