Gel Storage Method

Gel storage method is a laboratory procedure for preserving biological gels, such as agarose or polyacrylamide gels used in electrophoresis, so their separated DNA, RNA, or proteins remain available for imaging and analysis. The gel is typically kept hydrated in an appropriate buffer or sealed container under controlled temperature and light conditions, which limits drying, diffusion, microbial growth, and degradation of fluorescent or stained signals. Proper storage supports reliable documentation, band excision, downstream purification, and comparison of experimental results. In biology, this method helps researchers maintain sample integrity between electrophoresis, visualization, and subsequent molecular analyses.

Gel Purification

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...

A schema is a mental framework that helps individuals organize and interpret information. Schemata, formed from previous experiences, influence how we process new information: how we encode it, the inferences we make, and how we retrieve it. For instance, a schema for what a typical classroom looks like might include desks, a teacher's desk, a whiteboard, and students in such an environment. This expectation helps us quickly understand and navigate new classrooms without needing to analyze each...

Source: Robert M. Rioux & Taslima A. Zaman, Pennsylvania State University, University Park, PA While the use of various chemicals in experimental research is essential, it is also important to safely store and maintain them as a part of the Environmental, Health and Safety (EHS) program. The properties of chemicals and their reactivity vary broadly and if chemicals are not managed, stored, and labeled properly, they can have harmful or even destructive consequences such as toxic fume...
