Germplasm Collection Analysis

Germplasm collection analysis is the systematic evaluation of preserved genetic resources, such as seeds, plant tissues, or living specimens, to document their identity, diversity, and potential value in biology and crop research. Researchers combine passport data with morphological, physiological, and molecular characterizations to compare accessions, detect duplication, assess genetic relationships, and identify traits of interest. This analysis supports gene-bank management by guiding conservation priorities, improving collection organization, and revealing material useful for breeding, ecological studies, and responses to environmental stress. It also strengthens the responsible use of biological diversity in research and long-term conservation.

This protocol was developed to screen a large germplasm collection of the leafy vegetable lettuce (Lactuca sativa L.) for drought-tolerance, in order to identify a small candidate pool of lettuce for use in physiological, molecular, and genetic studies to identify underlying drought-tolerance traits along with breeding programs.

Blood Collection for Biochemical Analysis in Adult Zebrafish

Previous work suggests that the nitrogen isotopic composition of atmospheric nitrogen oxides might distinguish the influence of different sources in the environment. We report on an automated, mobile, field-based method for the high collection efficiency of atmospheric NOx for N isotopic analysis at an hourly time resolution.

Knowledge of the composition of the phloem sap as well as the mechanism of its loading and long-distance transport is essential for the understanding of long-distance signaling in plant development and stress/pathogen response and of assimilate transport. This manuscript describes the collection of phloem exudates using the EDTA-facilitated method.

A Psychophysics Paradigm for the Collection and Analysis of Similarity Judgments

The protocol presents an experimental psychophysics paradigm to obtain large quantities of similarity judgments, and an accompanying analysis workflow. The paradigm probes context effects and enables modeling of similarity data in terms of Euclidean spaces of at least five dimensions.
