Disease-relevant Cell Line

Disease-relevant cell lines are populations of cells maintained in vitro that reproduce key features of a human disease, providing accessible models for biological research. They are established from diseased tissues, patient samples, or genetically modified cells and cultured under controlled conditions that preserve disease-associated traits such as altered signaling, proliferation, or drug response. In biology, these models help researchers investigate disease mechanisms, compare healthy and diseased cell behavior, and evaluate potential therapies. Their reproducibility and experimental flexibility support studies of gene function, biomarker development, toxicity, and treatment resistance, while careful validation is essential to confirm that the cell line remains representative of the disease.

Longitudinal In Vivo Imaging of the Cerebrovasculature: Relevance to CNS Diseases

This manuscript describes a procedure to track the remodeling of the cerebrovasculature during amyloid plaque accumulation in vivo using longitudinal two-photon microscopy. A thinned-skull preparation enables the visualization of fluorescent dyes to assess the progression of cerebrovascular damage in a mouse model of Alzheimer's disease.

Protocol for propagation of dissociated high grade glioma surgical specimens in serum-free neurosphere medium to select for cells with cancer stem cell phenotype. For specimens that fail to grow as neurospheres, an alternative protocol is suggested. A paraffin embedding technique for maintaining the 3D neurosphere architecture for immunocytochemistry is described.

Hypoxia simulation in humans has usually been performed by inhaling hypoxic gas mixtures. For this study, apneic divers were used to simulate dynamic hypoxia in humans. Additionally, physiological changes in desaturation and re-saturation kinetics were evaluated with non-invasive tools such as Near-Infrared-Spectroscopy (NIRS) and peripheral oxygenation saturation (SpO2).

Here, we describe a protocol for engineering chemically reprogrammed stem cells to achieve precise neuronal modulation by differentiating these cells into dopaminergic precursor cells, transplanting them into mouse models of Parkinson's disease, and evaluating behavioral and electrophysiological outcomes to confirm the successful integration and functional effectiveness of the transplanted cells.
