Xeno-free Conditions

Xeno-free conditions are laboratory environments designed to maintain cells without components derived from nonhuman animals, reducing sources of biological variability and contamination. In cell culture, researchers replace animal-derived serum, feeder cells, enzymes, or matrix materials with defined recombinant, synthetic, or human-derived alternatives that supply nutrients, attachment signals, and growth factors under controlled conditions. These standardized systems support the expansion and differentiation of stem cells and other primary cells while improving reproducibility and compatibility with clinical manufacturing. Xeno-free culture is therefore important in developmental biology, disease modeling, tissue engineering, and the development of cell-based therapies.

The therapeutic potential of mesenchymal stem/stromal cells (MSCs) is well-documented, however the best method of preparing the cells for patients remains controversial. Herein, we communicate protocols to efficiently generate and administer therapeutic spherical aggregates or 'spheroids' of MSCs primed under xeno-free conditions for experimental and clinical applications.

This protocol describes the reprogramming of primary amniotic fluid and membrane mesenchymal stem cells into induced pluripotent stem cells using a non-integrating episomal approach in fully chemically defined conditions. Procedures of extraction, culture, reprogramming, and characterization of the resulting induced pluripotent stem cells by stringent methods are detailed.

Here, we present a protocol to precisely form hemogenic endothelium from human pluripotent stem cells in a feeder- and xeno-free condition. This method will have broad applications in disease modeling and screening for therapeutic compounds.

The production of specialized retinal cells from pluripotent stem cells is a turning point in the development of stem cell-based therapy for retinal diseases. The present paper describes a simple method for an efficient generation of retinal organoids and retinal pigmented epithelium for basic, translational, and clinical research.

A protocol is presented to prepare polyvinyl alcohol-co-itaconic acid hydrogels with varying stiffness, which were grafted with and without oligopeptides, to investigate the effect of the stiffness of biomaterials on the differentiation and proliferation of stem cells. The stiffness of the hydrogels was controlled by the crosslinking time.
