Goat Serum Blocking

Goat serum blocking is a laboratory technique that reduces nonspecific antibody binding before immunostaining or other antibody-based assays. The serum’s proteins bind exposed, nonspecific sites on cells, tissues, membranes, or assay surfaces, limiting unintended interactions while leaving target epitopes available for the primary antibody. Researchers select an appropriate serum concentration and incubation period based on the sample and detection system, often using serum from a species unrelated to the primary antibody host. This step lowers background signal, improves contrast and specificity, and supports more reliable results in immunohistochemistry, immunofluorescence, Western blotting, and related biological analyses.

Preimplantation embryos may be cryopreserved after placement into a hypertonic cryoprotective solution to cause cellular dehydration. After equilibration, ice crystal formation is induced in the solution surrounding the embryo. Further dehydration occurs as the embryo is slowly cooled to subzero temperatures before plunging into liquid nitrogen for storage.

This protocol describes a method for extracting small RNAs from human serum. We have used this method to isolate microRNAs from cancer serum for use in DNA arrays and also singleplex quantitative PCR. The protocol utilizes phenol and guanidinium thiocyanate reagents with modifications to yield high quality RNA.

An Assay to Detect Autoantibodies in Human Serum using a Hippocampal Neuronal Culture

2025

This video demonstrates a technique for identifying antibodies against specific antigens in a sample using a rat hippocampal neuronal culture. By exposing cultured neurons to patient samples containing autoantibodies targeting N-Methyl-D-aspartate receptors (NMDARs), the presence of autoantibodies bound to the neuronal surface is revealed through immunostaining.
