Graded Potentials

Graded potentials are variable changes in a cell’s membrane potential that enable neurons and other excitable cells to respond to stimuli. They arise when stimulus-gated or ligand-gated ion channels open, altering the movement of ions across the membrane and producing depolarization or hyperpolarization; their amplitude depends on stimulus strength, and the signal weakens as it spreads from its origin. In nervous tissue, graded potentials occur in dendrites, cell bodies, and sensory receptor endings, where they integrate synaptic inputs and sensory information. If depolarization reaches threshold at the axon hillock, it can trigger an all-or-none action potential, supporting rapid communication throughout the nervous system.

This video showcases a technique for co-culturing glutamatergic neurons and pediatric high-grade glioma (pHGG) cells within a microfluidic device. Human induced pluripotent stem cells are introduced into the microfluidic system and incubated to promote their differentiation into mature cortical glutamatergic neurons. Following this, the pHGG cells are introduced onto the mature neurons, establishing a co-culture and enabling interactions between neurons and pHGG cells.
