Gst-rho Binding Domain

The GST-Rho binding domain is a recombinant protein construct used to detect and isolate activated Rho-family GTPases, key regulators of cytoskeletal organization, cell adhesion, and migration. It typically combines glutathione S-transferase (GST) with a Rho-binding domain from an effector protein; the domain selectively binds the GTP-loaded conformation of Rho proteins, while inactive GDP-bound forms show little or no interaction. In GST pull-down assays, the fusion protein is immobilized on glutathione-coated beads and incubated with cell lysates, allowing active Rho to be recovered and measured by immunoblotting or related analyses. This approach supports studies of Rho signaling, cellular dynamics, and disease-associated pathway changes.

Affinity Precipitation of Active Rho-GEFs Using a GST-tagged Mutant Rho Protein (GST-RhoA(G17A)) from Epithelial Cell Lysates

The method presented here describes an assay to follow activation of RhoA specific GDP/GTP Exchange Factors (GEFs) in cultured cells by making use of a mutant RhoA GST fusion protein that has high affinity for activated GEFs. GEFs are precipitated from cell lysates, detected by Western blotting and quantified by densitometry.

In the present protocol, we demonstrate a highly efficient and cost-effective small-scale protein purification method, which allows purification of recombinant proteins by uniquely combining a cleavable GST-tag and a small His-tag.

Self-assembled polyelectrolyte complexes (PEC) fabricated from heparin and protamine were deposited on alginate beads to entrap and regulate the release of osteogenic growth factors. This delivery strategy enables a 20-fold reduction of BMP-2 dose in spinal fusion applications. This article illustrates the benefits and fabrication of PECs.

A method for large-scale purification of the APP intracellular domain (AICD) is described. We also describe methodology to induce in vitro AICD aggregation and visualization by atomic force microscopy. The methods described are useful for biochemical/structural characterization of the AICD and the effects of molecular chaperones on its aggregation.
