Headspace Techniques

Headspace techniques are analytical methods that examine volatile compounds in the gas phase above a solid or liquid sample, allowing biological materials to be analyzed with minimal direct handling. In a sealed vial, compounds partition between the sample and its surrounding air until they approach equilibrium; an aliquot of the headspace is then withdrawn manually or with an automated sampler and commonly introduced into gas chromatography for separation and detection. In biology, these methods help measure volatile metabolites, odors, fermentation products, and gases released by cells, tissues, foods, or microorganisms. They support studies of metabolism, microbial activity, environmental responses, and sample quality while reducing contamination and matrix-related interference.
