Hml Hmr Loci

HML and HMR loci are silent mating-type cassettes in the budding yeast Saccharomyces cerevisiae, providing stored genetic information that supports controlled mating-type switching. Specialized silencer sequences recruit Sir proteins and associated chromatin factors, forming a repressive chromatin environment that prevents transcription, while the HO endonuclease initiates switching at the active MAT locus and DNA repair copies information from HML or HMR through gene conversion. These loci offer a well-defined model for studying epigenetic gene silencing, chromatin organization, DNA double-strand break repair, and inheritance of transcriptional states, with broader relevance to mechanisms that regulate genome stability and cell identity.

2013

Human endogenous retroviruses (HERV), which occupy 8% of the human genome, retain scarce coding capacities but a hundred thousand long terminal repeats (LTRs). A custom Affymetrix microarray was designed to identify individual HERV locus expression and was used on prostate cancer tissues as a proof of concept for future clinical studies.

Root Loci for Positive-Feedback Systems

2024

The Hartley oscillator is a positive feedback system that sustains oscillations by feeding the output back to the input in phase, thereby reinforcing the signal. Positive feedback systems can be viewed as negative feedback systems with inverted feedback signals. In these systems, the root locus encompasses all points on the s-plane where the angle of the system transfer function equals 360 degrees. The construction rules for the root locus in positive feedback systems are similar to those in...

Combining DNA halo preparations with fluorescence in situ hybridization enables high-resolution analysis of genomic interactions with the nucleoskeleton. Attached genome leads to hybridized fluorescent signals located within the residual extracted nuclei, whereas non-attached genome is in the halo of DNA surrounding the residual nuclei.

HOX Loci Focused CRISPR/sgRNA Library Screening Identifying Critical CTCF Boundaries

A CRISPR/sgRNA library has been applied to interrogating protein-coding genes. However, the feasibility of a sgRNA library to uncover the function of a CTCF boundary in gene regulation remains unexplored. Here, we describe a HOX loci specific sgRNA library to elucidate the function of CTCF boundaries in HOX loci.
