Host-virus Interaction

Host-virus interaction is the dynamic relationship between a virus and the infected organism, determining whether infection is established, controlled, or spread. Viral surface proteins recognize specific host-cell receptors to enable attachment and entry, after which the virus uses cellular machinery for replication while host innate and adaptive immune responses detect and counter the infection; viruses may also evolve mechanisms that evade these defenses. Studying these interactions clarifies viral pathogenesis, tissue tropism, and disease progression. It also supports the development of antiviral drugs, vaccines, diagnostics, and strategies for predicting how viral mutations may alter transmission or host range.

Dissecting Host-virus Interaction in Lytic Replication of a Model Herpesvirus

We describe a protocol to identify key roles of host signaling molecules in lytic replication of a model herpesvirus, gamma herpesvirus 68 (γHV68). Utilizing genetically modified mouse strains and embryonic fibroblasts for γHV68 lytic replication, the protocol permits both phenotypic characterization and molecular interrogation of virus-host interactions in viral lytic replication.

Intrathoracic Viral Nano-Injection to Study Host-Virus Interactions in Adult Flies

In this video, we describe a procedure to perform a nano-injection of a positive-sense, single-stranded RNA virus into adult male Drosophila flies and subsequently study the antiviral responses initiated due to the infection. Polymerase chain reaction analysis identifies the upregulation of specific genes in the infected flies that indicate the activation of immune response pathways including RNA interference and JAK/STAT signaling.

This protocol describes the generation of polymer-coupled recombinant adhesins as biomimetic materials which allow analysis of the contribution of individual adhesins to these processes, independent of other bacterial factors.
