Ifn-gamma Resistance

IFN-gamma resistance is the impaired ability of cells or tissues to respond to interferon-gamma, a cytokine that coordinates antimicrobial immunity and regulates inflammation. Normally, interferon-gamma binds its receptor and activates the JAK-STAT signaling pathway, enabling STAT1-dependent gene expression; defects in IFNGR1, IFNGR2, STAT1, or related signaling components can weaken this response. In biology, studying IFN-gamma resistance helps explain susceptibility to intracellular pathogens, particularly mycobacteria, and reveals how immune signaling fails in genetic immunodeficiencies, chronic disease, or tumors. Characterizing these defects can guide diagnosis, clarify host-pathogen interactions, and support development of targeted therapies that restore immune function.

Assay for the Identification of Interferon-Gamma Secretion in Murine Lymphoid Organs

This video demonstrates an imaging method to visualize in situ interferon-gamma (IFN-γ) secretion in the mouse spleen. Following bacterial infection and the induction of IFN-γ accumulation inside T cells and natural killer cells, the spleen is harvested from the mouse. Upon obtaining tissue sections, immunostaining is performed to visualize IFN-γ inside the cells.
