Immunofluorescence Sample Prep

Immunofluorescence sample prep comprises the steps used to preserve biological specimens and make cellular targets accessible for fluorescent antibody labeling and microscopy. Fixation stabilizes cell and tissue structure, while permeabilization allows antibodies to reach intracellular antigens; blocking reagents reduce nonspecific binding before primary and fluorescently tagged secondary antibodies are applied. Careful preparation helps maintain morphology, preserve antigen availability, and limit background signal, producing clearer localization of proteins and other biomolecules. In biology, these methods support studies of cell organization, protein expression, tissue architecture, disease-associated changes, and molecular interactions in cultured cells or tissue sections.

Here we describe a method to quantify molecular heterogeneity in histological sections of tumor material using quantitative immunofluorescence, image analysis, and a statistical measure of heterogeneity. The method is intended for use in clinical biomarker development and analysis.

Here we present a robust immunofluorescence and image analysis method to assess cardiomyocyte-specific structures in the developing mouse heart.
