Inhalation Anesthesia

Inhalation anesthesia is a method of delivering volatile anesthetic agents through the respiratory tract to produce reversible unconsciousness and reduced sensation during medical or surgical procedures. These agents enter the alveoli, diffuse into the pulmonary blood, and reach the brain, where their concentration-dependent effects depress neuronal activity; adjusting inspired concentration and ventilation helps control anesthetic depth. In biology and medicine, inhalation anesthesia supports controlled experiments involving animals and enables surgery while physiological variables can be monitored and managed. Understanding uptake, distribution, metabolism, and elimination helps researchers interpret behavioral and physiological outcomes, reduce complications, and select protocols suited to the organism and procedure.

Here, we describe two novel methods of stable intranasal administration under inhalation anesthesia with minimal physical stress for experimental animals. We also describe a method for quantitative evaluation of drug distribution levels in the brain via the nose-to-brain pathway using radiolabeled [14C]-inulin as a model substrate of water-soluble macromolecules.

A whole-body nanoparticle aerosol inhalation exposure facility was constructed for nano-sized titanium dioxide (TiO2) inhalation toxicology studies. This system provides nano-TiO2 aerosol test atmospheres that have: 1) a steady mass concentration; 2) a homogenous composition free of contaminants; and 3) a stable particle size distribution during aerosol generation.
