Insertion Sequences

Insertion sequences (IS elements) are short, mobile DNA segments that move within and between bacterial genomes, shaping genome structure and genetic variation. Their transposase recognizes inverted repeat sequences at the element’s ends and catalyzes insertion into a new DNA site, often producing short target-site duplications; depending on the element, movement can follow cut-and-paste or replicative pathways. Insertion sequences can disrupt genes, alter transcription, rearrange chromosomes, and mobilize neighboring DNA, including antibiotic-resistance determinants. Studying IS elements therefore helps explain bacterial adaptation, genome evolution, and the spread of traits relevant to microbiology and biotechnology.

Generating Transposon Insertion Libraries in Gram-Negative Bacteria for High-Throughput Sequencing

We describe a method to generate saturating transposon mutant libraries in Gram-negative bacteria and subsequent preparation of DNA amplicon libraries for high-throughput sequencing. As an example, we focus on the ESKAPE pathogen, Acinetobacter baumannii, but this protocol is amenable to a wide range of Gram-negative organisms.

Transposon-insertion Sequencing as a Tool to Elucidate Bacterial Colonization Factors in a Burkholderia gladioli Symbiont of Lagria villosa Beetles

This is an adapted method for identifying candidate insect colonization factors in a Burkholderia beneficial symbiont. The beetle host is infected with a random mutant library generated via transposon mutagenesis, and library complexity after colonization is compared to a control grown in vitro.
