Interstitial Fluid Collection

Interstitial fluid collection is the process of obtaining the extracellular fluid that surrounds tissue cells, where it supports nutrient exchange, waste removal, and cell signaling. This fluid originates largely from capillary filtration, moves through the extracellular matrix, and normally returns to the circulation through lymphatic drainage; collection methods sample it directly from tissue spaces for analysis. Measuring its composition can reveal local concentrations of metabolites, proteins, inflammatory mediators, and therapeutic compounds that may differ from blood. In biology and biomedical research, interstitial fluid collection therefore supports studies of tissue physiology, disease mechanisms, biomarker distribution, and drug delivery.

Interstitial fluid flow is elevated in solid tumors and can modulate tumor cell invasion. Here we describe a technique to apply interstitial fluid flow to cells embedded in a matrix and then measure its effects on cell invasion. This technique can be easily adapted to study other systems.
