Live Cells

Live cells are intact, metabolically active cells that maintain membrane integrity, generate energy, and respond to their surroundings, making them essential for studying biology as it occurs in real time. Their continued activity depends on coordinated processes such as nutrient uptake, cellular respiration, signaling, growth, division, and homeostatic regulation under suitable environmental conditions. Researchers examine live cells using cell culture, microscopy, viability assays, and fluorescent reporters to track morphology, movement, intracellular dynamics, and responses to stimuli or treatments. These approaches support investigations of development, disease mechanisms, drug effects, and tissue function while preserving dynamic cellular behavior that fixed or isolated components cannot reveal.

4D Imaging of Protein Aggregation in Live Cells

Cellular viability depends on timely and efficient management of protein misfolding. Here we describe a method for visualizing the different potential fates of a misfolded protein: refolding, degradation, or sequestration in inclusions. We demonstrate the use of a folding sensor, Ubc9ts, for monitoring proteostasis and aggregation quality control in live cells using 4D microscopy.

Cell Death Associated with Abnormal Mitosis Observed by Confocal Imaging in Live Cancer Cells

The cytotoxic activity of the phenanthridine PJ-34 in cancer cells undergoing mitosis was documented in real time by live confocal imaging. PJ-34 eradicated human breast cancer MDA-MB-231 cells harboring extra-centrosomes in mitosis. Unlike normal bi-focal mitosis, the extra-centrosomes were not clustered in the two spindle poles in the presence of PJ-34.
