Live Confocal Microscopy

Live confocal microscopy is an imaging technique that captures high-resolution, optical-sectioned views of living cells, tissues, and organisms over time. A focused laser scans fluorescently labeled specimens, while a pinhole blocks out-of-focus light so the detector records sharp images from selected depths; sequential scans can then be assembled into time-lapse sequences. In biology, this approach reveals dynamic processes such as cell migration, organelle movement, signaling, and tissue development without requiring fixed specimens. Its ability to combine spatial detail with temporal information supports studies of cellular behavior, disease mechanisms, and responses to experimental treatments.

In this video, we demonstrate a method by which to analyze the developing vertebrate brain in live zebrafish embryos at single cell resolution by confocal microscopy. This includes the method by which we inject the single-cell zebrafish embryo and subsequently mount and image the developing brain.

Several methods are available for measuring intracellular pH but very few of these allow simultaneous measurement of cytoplasmic and organellar pH. Here, we describe in detail a rapid and accurate methodology to simultaneously measure cytoplasmic and vesicular pH by ratiometric imaging of living cells.
