Liver Dysfunction

Liver dysfunction is the impaired ability of the liver to perform essential functions such as metabolism, detoxification, bile production, and synthesis of blood proteins. It develops when injury or disease disrupts hepatocyte activity, reducing the organ’s capacity to process nutrients and medications, remove harmful compounds, regulate glucose and lipid balance, or produce proteins involved in blood clotting. In biology and biomedical research, studying liver dysfunction helps explain conditions ranging from inflammation and fibrosis to cirrhosis and organ failure. Measures of liver enzymes, bilirubin, and synthetic function support assessment of injury, while experimental models aid research into disease mechanisms, therapeutic targets, and liver regeneration.

Mouse model of metabolic dysfunction-associated steatotic liver disease (MASLD) with metabolic dysfunction, hepatic gene expression changes, and liver histopathological alterations that resemble human MASLD, including fibrosis that progresses to advanced fibrosis stage 3. Here we present a protocol describing oxygenated ex situ machine perfusion of donor liver grafts. This article contains a step by step protocol to procure and prepare the liver graft for machine perfusion, prepare the perfusion fluid, prime the perfusion machine and perform oxygenated normothermic machine perfusion of the liver graft.

Decellularization and Recellularization of Whole Livers

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.
