Matrigel Scaffold

A Matrigel scaffold is a three-dimensional, basement membrane-like matrix used to support cells in laboratory culture, helping researchers model tissue organization and function. Derived from mouse tumor extracellular matrix, Matrigel contains structural proteins such as laminin and collagen IV and forms a gel under physiological temperature conditions, providing adhesion sites and biochemical cues that influence cell survival, growth, migration, and differentiation. In biology, it supports organoid formation, stem cell culture, tissue modeling, and assays of development or disease. Matrigel-based systems can better reproduce aspects of the cellular microenvironment than two-dimensional cultures, although composition variability can affect experimental reproducibility.

This video demonstrates how to maintain the growth of human embryonic stem cells (hESCs) in feeder cell-free conditions and how to continuously passage hESCs in feeder cell-free conditions. Confirmation of hESC pluripotency grown in feeder cell-free conditions by immunofluorescence microscopy is also demonstrated. Part 3 of 3.
