Melanin Production

Melanin production is the biological process by which specialized cells synthesize melanin pigments, which help determine pigmentation and protect tissues from ultraviolet radiation. In melanocytes, the enzyme tyrosinase converts the amino acid tyrosine through a series of reactions into melanin within cellular organelles called melanosomes; these pigment-containing structures are then transferred to neighboring skin cells. In biology, studying this process clarifies how genetic, hormonal, and environmental factors influence skin, hair, and eye color. It also supports research into pigmentation disorders, UV-induced cellular damage, melanoma, and potential treatments that regulate pigment formation.

Epidermal melanin is induced by topical application of forskolin in a murine model of the fair-skinned UV-sensitive human. Pharmacologic manipulation of cAMP levels in the skin and epidermal darkening strongly protect against UV-mediated inflammation (sunburn) as measured by the minimum erythematous dose (MED) assay.

Bacterial pyomelanin production results in increased resistance to oxidative stress and virulence. We report on techniques that can be used to determine inhibition of pyomelanin production and assay the resulting increase in sensitivity to oxidative stress in bacteria, as well as determine antibiotic minimum inhibitory concentration (MIC).
