Mitotic Entry Analysis

Mitotic entry analysis is the study of how and when a cell transitions from interphase into mitosis, a key event for understanding cell-cycle control and accurate chromosome segregation. This transition occurs when cyclin-dependent kinase 1 (CDK1) activates in complex with cyclin B, triggering chromosome condensation, centrosome maturation, and nuclear envelope breakdown; researchers assess these events using microscopy, DNA-content measurements, and mitotic markers such as phospho-histone H3. In biology, mitotic entry analysis helps characterize cell proliferation, checkpoint function, developmental timing, and responses to genetic or chemical perturbations. It also supports research into cancer biology, where disrupted cell-cycle regulation can produce uncontrolled division and genomic instability.

Mitosis is critical to every living organism and defects often lead to cancer and developmental disorders. Using this imaging protocol and zebrafish as a model system, researchers can visualize mitosis in a live vertebrate organism and the multitude of defects that arise when mitotic processes are defective.

Among the three mosquito genera, namely Anopheles, Aedes, and Culex, physical genome mapping techniques were established only for Anopheles, whose members possess readable polytene chromosomes. For the genera of Aedes and Culex, however, cytogenetic mapping remains challenging because of the poor quality of polytene chromosomes. Here we present a universal protocol for obtaining high-quality preparations of mitotic chromosomes and an optimized FISH protocol for all three genera of mosquitoes.
