Mouse Lens Stiffness

Mouse lens stiffness is the biomechanical resistance of the eye’s crystalline lens to deformation, a property that helps determine how the lens changes shape and focuses light. It reflects the organization and material behavior of densely packed lens fiber cells, their crystallin-rich cytoplasm, and the lens’s hydration state, and can be assessed through mechanical deformation measurements. Studying stiffness in mice supports research on lens development, aging, accommodation, and cataract formation by linking changes in tissue mechanics with optical performance. These measurements also provide a model for understanding how altered lens biomechanics contribute to visual dysfunction.

Age-related increases in eye lens stiffness are linked to presbyopia. This protocol describes a simple, cost-effective method for measuring mouse lens stiffness. Mouse lenses, like human lenses, become stiffer with age. This method is precise and can be adapted for lenses from larger animals.

Developing Myopia in a Mouse Model Through Lens-Induced Defocusing

Source: Jiang, X., et al. Inducement and evaluation of a murine model of experimental myopia. J. Vis. Exp. (2019)This video demonstrates a procedure for inducing myopia in a mouse through the installation of a custom lens frame. The frame is secured to the exposed skull with dental adhesive, and concave lenses are inserted to shift the focal point behind the retina. This setup stimulates eyeball elongation leading to myopia.

This protocol details the separation of the mouse ocular lens epithelium and fiber cell bulk mass, followed by RNA isolation and qPCR analysis. This method allows for separating lens cell compartments for a more detailed analysis of the transcriptome and biological processes in epithelial cells vs. differentiated fiber cells.
