Fungal Aryl-alcohol Oxidase

Fungal aryl-alcohol oxidase is a flavin-dependent enzyme that oxidizes aromatic alcohols and contributes to the breakdown of lignin-rich plant material. It transfers electrons from an aryl alcohol to molecular oxygen, converting the alcohol into its corresponding aromatic aldehyde while generating hydrogen peroxide as a by-product. In wood-decaying fungi, this peroxide can support other oxidative enzymes involved in lignocellulose degradation, helping organisms access carbon in woody biomass. Studying aryl-alcohol oxidase clarifies fungal decomposition and oxidative metabolism, while its ability to produce valuable aldehydes and hydrogen peroxide supports potential applications in biotechnology, biocatalysis, and biomass processing.

NADPH oxidase is the major source of reactive oxygen species (ROS) in phagocytes. Because of the ephemeral nature of ROS, it is difficult to measure and monitor ROS levels in living animals. A minimally invasive method for serial quantification of ROS in living mice is described.

We describe methods for live-cell video microscopy of Candida albicans phagocytosis by macrophages. These methods enable stage-specific analysis of macrophage migration, recognition, engulfment and phagosome maturation and reveal novel aspects of phagocytosis.
