Mrna Transcription Kinetics

mRNA transcription kinetics describes the time-dependent rates and dynamics of messenger RNA production from DNA, revealing how cells regulate gene expression. RNA polymerase initiates transcription at a promoter, elongates the RNA strand by reading the DNA template, and terminates at defined signals; measurements can distinguish initiation frequency, elongation rate, and transcript lifetime. In biology, kinetic analysis helps explain how regulatory proteins, chromatin state, and cellular conditions alter gene activity. These measurements support studies of development, stress responses, disease mechanisms, and drug effects by linking molecular events to changes in mRNA abundance over time.

This method describes high yield in vitro synthesis of both capped and uncapped mRNA from a linearized plasmid containing the Gaussia luciferase (GLuc) gene. The RNA is purified and a fraction of the uncapped RNA is enzymatically capped using the Vaccinia virus capping enzyme. In the final step, the mRNA is transfected into HeLa cells and cell culture supernatants are assayed for luciferase activity.

Nuclear Export of mRNA

2020

Before mRNAs are exported to the cytoplasm, it is crucial to check each mRNA for structural and functional integrity. Eukaryotic cells use several different mechanisms, collectively known as mRNA surveillance, to look for irregularities in mRNAs. Irregular or aberrant mRNA are rapidly degraded by various enzymes. If a defective mRNA escapes the surveillance, it would be translated into a protein which would either be non-functional or not function properly. One of the primary irregularities in...
