Neuron Membrane

The neuron membrane is a selectively permeable lipid bilayer that surrounds nerve cells and regulates communication between the neuron and its environment. Membrane proteins, including ion channels, pumps, and receptors, control the movement of charged particles such as sodium and potassium, maintaining an electrochemical gradient across the membrane. When stimulation reaches threshold, coordinated ion flow rapidly changes the membrane potential and generates an action potential that travels along the axon. Understanding neuron membrane structure and signaling is essential for studying sensory processing, nervous system function, neurological disorders, and the effects of drugs that alter ion channels or receptor activity.

The neuronal membrane proteasome (NMP) was recently identified in a subset of somatosensory neurons as a key regulator of touch, pain, and itch perception. This article presents a robust workflow for analyzing cell-type-specific NMP expression and function using cell sorting, transcriptome profiling, and culture-based immunofluorescent analyses.

Co-Culture of Schwann Cells with DRG Neurons on a Pre-Stretched Membrane

The video demonstrates the co-culturing of dorsal root ganglion (DRG) neurons with Schwann cells. DRG neurons are first grown on a pre-stretched membrane for aligned axon growth. Schwann cells are then introduced, which attach to the axons, insulating them and promoting neural cell survival and function.

Imaging pHluorin-tagged Receptor Insertion to the Plasma Membrane in Primary Cultured Mouse Neurons

By tagging the extracellular domains of membrane receptors with superecliptic pHluorin, and by imaging these fusion receptors in cultured mouse neurons, we can directly visualize individual vesicular insertion events of the receptors to the plasma membrane. This technique will be instrumental in elucidating the molecular mechanisms governing receptor insertion to the plasma membrane.
