Neuronal Transport

Neuronal transport is the movement of proteins, vesicles, organelles, and other cargo within neurons, a process essential for maintaining long axons and supporting communication between cells. Molecular motor proteins, including kinesin and dynein, carry cargo along polarized microtubules through anterograde transport from the cell body toward axon terminals and retrograde transport back to the cell body. This system delivers synaptic components, removes damaged materials, and enables signaling between distant neuronal regions. Studying neuronal transport helps explain axonal growth, synaptic function, and neurodegenerative diseases in which transport pathways become disrupted.

Neuromodulation and Mitochondrial Transport: Live Imaging in Hippocampal Neurons over Long Durations

We describe a protocol that allows imaging of mitochondria in living neurons via fluorescence microscopy over long durations. Imaging over extended periods is accomplished through lentivirus-mediated expression of a mitochondrially targeted fluorescent protein and use of an inexpensive stage-top incubator that was designed and built in our laboratory.

This video demonstrates a method for tracking axonal transport in motor neurons using a microfluidic chamber system. It involves culturing mouse embryonic spinal cord tissues in polymer-coated wells with nutrient medium to induce axonal growth. The movement of fluorescent dye-stained organelles in live imaging, confirms the bidirectional axonal transport.
