Noncoding Rna Annotation

Noncoding RNA annotation is the process of identifying, cataloging, and characterizing RNA transcripts that do not primarily encode proteins, helping researchers interpret functional elements within genomes. It combines sequence analysis with transcriptome data, genomic-location mapping, evolutionary conservation, and structural or expression evidence to distinguish noncoding RNAs from protein-coding transcripts and assign features or probable functions. In biology, annotation supports the study of regulatory RNAs, genome organization, development, and disease-associated changes. Accurate catalogs also improve genome assemblies, guide experimental validation, and provide a foundation for comparing noncoding RNA repertoires across cells, tissues, species, and biological conditions.

Isolation of Small Noncoding RNAs from Human Serum

This protocol describes a method for extracting small RNAs from human serum. We have used this method to isolate microRNAs from cancer serum for use in DNA arrays and also singleplex quantitative PCR. The protocol utilizes phenol and guanidinium thiocyanate reagents with modifications to yield high quality RNA.

Genome Annotation and Assembly

The genome refers to all of the genetic material in an organism. It can range from a few million base pairs in microbial cells to several billion base pairs in many eukaryotic organisms. Genome assembly refers to the process of taking the DNA sequencing data and putting it all back together in a correct order to create a close representation of the original genome. This is followed by the identification of functional elements on the newly assembled genome, a process called genome annotation.
