Nuclei Lysis

Nuclei lysis is the controlled disruption of cell nuclei to release genomic DNA, chromatin, RNA, and nuclear proteins for biological analysis. It typically uses hypotonic buffers, detergents, enzymatic treatment, or mechanical shearing to weaken and rupture the nuclear envelope while preserving the molecules of interest; conditions are adjusted to limit degradation and unwanted contamination. In biology, nuclei lysis supports DNA and chromatin extraction, nucleic acid purification, protein analysis, and preparation of samples for sequencing or molecular assays. Effective lysis improves recovery and reproducibility, making it a critical step in studies of gene regulation, genome organization, and cellular function.

This video describes the protocol to isolate single-nuclei from fresh frozen glioma tissue. The isolated single-nuclei can be used as specimens for sequencing techniques such as single-nuclei RNA (snRNA) sequencing and single-nuclei Assay for Transposase-Accessible Chromatin (snATAC) sequencing to obtain deeper insights into inter- and intra-tumor heterogeneity observed in glioma of the central nervous tissues.
