Onion Epidermal Cells

Onion epidermal cells are a thin, transparent layer of plant cells lining an onion bulb, commonly used as a simple model for studying cell structure and transport in biology. Each cell has a cellulose cell wall, plasma membrane, cytoplasm, nucleus, and a large central vacuole, making major structures visible with light microscopy after staining. In a hypertonic solution, water leaves the cell by osmosis, causing the membrane and cytoplasm to pull away from the wall, a process called plasmolysis. Observing these cells helps students and researchers investigate plant anatomy, microscopy, membrane behavior, and the effects of solute concentration.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.
